Inhibition of Cytochrome P450 by Carbon Monoxide: Relevance to Drug Resistance in Human Breast Cancer Therapy
نویسندگان
چکیده
Drug resistance to conventional chemotherapeutics is a great impediment cancer therapy. A major part of this problem arises from rapid metabolism the drugs by cytochrome P450 class enzymes before they reach their targets or at target itself. Inhibition such enzymatic deactivation could offer partial rescue and make chemotherapy more effective. Site specific delivery exogenous carbon monoxide has been shown inhibit resurrect sensitivity already available in market. Successful design for application CO will thus be extremely desirable patients particularly poor countries where antibody-based nanodrug therapies, discovered recently, are too expensive general population. The potential monoxide-induced inhibition improve drug sensitization breast therapy discussed account.

Inhibition of human drug metabolizing cytochrome P450 by buprenorphine.
The effects of buprenorphine, a powerful mixed agonist/antagonist analgesic, on several cytochrome P450 (CYP) isoform specific reactions in human liver microsomes were investigated to predict drug interaction of buprenorphine in vivo from in vitro data. The following eight CYP-catalytic reactions were used in this study: CYPlA1/2-mediated 7-ethoxyresorufin O-deethylation, CYP2A6-mediated coumar...
متن کاملElectrochemical Detection of Anti-Breast-Cancer Agents in Human Serum by Cytochrome P450-Coated Carbon Nanotubes
We report on the electrochemical detection of anti-cancer drugs in human serum with sensitivity values in the range of 8-925 nA/μM. Multi-walled carbon nanotubes were functionalized with three different cytochrome P450 isoforms (CYP1A2, CYP2B6, and CYP3A4). A model used to effectively describe the cytochrome P450 deposition onto carbon nanotubes was confirmed by Monte Carlo simulations. Voltamm...
متن کاملDrug-enhanced carbon monoxide production from heme by cytochrome P450 reductase
Carbon monoxide (CO) formed endogenously is considered to be cytoprotective, and the vast majority of CO formation is attributed to the degradation of heme by heme oxygenases-1 and -2 (HO-1, HO-2). Previously, we observed that brain microsomes containing HO-2 produced many-fold more CO in the presence of menadione and its congeners; herein we explored these observations further. We determined t...
متن کاملInhibition of Cytochrome P450 by Propolis in Human Liver Microsomes
Although propolis is one of the most popular functional foods for human health, there have been no comprehensive studies of herb-drug interactions through cytochrome P450 (CYP) inhibition. The purpose of this study was to determine the inhibitory effects of propolis on the activities of CYP1A2, 2A6, 2B6, 2C9, 2C19, 2D6, 2E1 and 3A4 using pooled human liver microsomes (HLMs). Propolis inhibited ...
متن کاملCytochrome p450 and glutathione transferase expression in human breast cancer.
PURPOSE The cytochrome P-450 (CYP) and glutathione S-transferase (GST) enzyme systems may influence the biological effects of carcinogens, including estrogens. As such, these enzymes may predict the developmental risk of breast cancer, as well as be potential targets for chemoprevention. The purpose of this study was to compare the expression of GST-Pi and CYPs 1A1, 2B6, 2E1, and 3A4 in paired ...
